Abstract

In this presentation, two different types of bioreducible inorganic nanoparticles releasing metal ions without cytotoxicity will be introduced. Mn ion at high concentration can be delivered to PC12 cells using AuNPs to enhance cellular neurodevelopment without causing cell death. Also, Fe ion at high concentrations can be delivered to hMSCs using AuNPs to improve angiogenic growth factors without inducing cell damage. Ion release from AuNPs was designed to be pH-sensitive so that low pH condition of the cell endosome can trigger in situ release of ions from AuNPs after cellular uptake of Mn- or Fe-incorporated AuNPs. Compared to PC12 cells treated with a high concentration of free Mn ion, PC12 cells treated with an equal concentration of MnAuNPs resulted in significantly enhanced cellular neurodevelopment with decreased apoptosis and necrosis. Treating AuFeNPs to hMSCs led enhanced therapeutic effect on skin wound healing based on improved angiogenic paracrine factor secretion.
